Plants have been recognized as major sources of an extremely wide range of organic chemicals. Many compounds of the plant origin are used in food flavorings and colorings, in perfumery, and specially provide the basis for medicine. The monoterpene perillyl alcohol and limonene are natural product mainly from orange, and they have chemotherapeutic activity against chemically induced rat mammary tumors with little toxicity to the host. In addition, perillyl alcohol has chemopreventive activity against rat liver cancer, and it inhibits the proliferation of cultured human colon carcinoma cell. Perillyl alcohol and d-limonene are currently undergoing clinical testing in cancer patients. RP-HPLC was applied to purity perillyl alcohol from Korean orange peel. The extract was obtained by solvent. Separation were achieved on a μBondapak C18 (3.9x300mm, 10μm) column and UV wavelength was fixed at 205nm. Identification of perillyl alcohol was confirmed by the standard from Aldrich. The mobile phase for isolating perillyl alcohol from the extract consisted of water/acetonitrile, 70/30(vol.%) and water/methanol/acetonitrile, 60/20/20(vol.%). The retention time of the completely resolved perillyl alcohol was 32 min in the two mobile phases.
