Evaluation of spinal fusion using 3D cultured BM-derived mesenchymal stem cells in rat

PURPOSE : We investigated the effectiveness of graft material for spinal fusion using 3D cultured BM mesenchymal stem cells cell in collagen sponge. MATERIALS AND METHODS : Twenty-four mature male SD rats (weight, 320-400g) were divided into 4 groups: Group I, collagen sponge only Group II, CS + MSC (1×106) Group III, 3D collagen cultured MSC(1×106) Group IV, CS + MSC (1×107). Each group underwent fusion of the intertransverse processes. The lumbar spine was harvested en bloc, and the fusion mass was evaluated radiographically, by manual palpation test, and by histologic analysis at 8 weeks after surgery. RESULTS : Fusion success or failure was assumed based on the results from manual palpation of the harvested spine. Four of 6 rats in the group II and group III, 5 of 6 rats in group IV and none of 6 rats in the collagen only group achieved fusion in the radiological and histological study. CONCLUSION : The present study suggested that MSCs that have been three dimentional cultured with collagen was not better in the formation of new bone in experimental spinal fusion than collagen injected MSC cells. However, suitable bone fusion could be confirmed to depend on injected cell number.
