Control of Fc Glycan Structures through Supplementation of Hexose Monosaccharide in CHO Cell Cultures

Glycosylation shows a significant impact on protein structure and function, and the biological functions ofantibodies rely on their oligosaccharide structures. Many studies have been focused on regulating glycans toimprove the quality of biopharmaceuticals [1]. The amount of available carbon sources causes the differences such as macroheterogeneity and microheterogeneity of glycans, because nucleotide sugars are synthesized from carbon sources in culture medium [2]. In this study, we investigated the effects of hexose monosaccharide ratios (glucose: galactose: mannose) on Fc glycans. Since nucleotide sugars interact with each other, addition of mixed carbon sources can lead to statistical change in glycosylation pattern, compared to those with separate addition. The hexoses were added in multiple conditions according to a central composite design using a Minitab. as results of hexose feeding, it was found that there was a tendency of glycans on Fc region by using different ratios of carbon sources. In addition, composition of hexose monosaccharide ratios could be optimized. In conclusion, various concentrations of sugars in media can shift glycosylation profiles, and an optimal culture medium composition can be used to control glycans for targeted structures.
