Upgrade of the vector efficiency during the production of gene manipulated mice models

Gene-manipulated animal model system can be characterized into gain-of-function and loss-of-function, which are called as transgenic and knock-out studies, respectively. Since it was uncovered for the first time about third decade ago, numerous sophisticated technologies have been developed and approached to upgrade the components including the components of vector such as selection marker. To make knock-out mice, it needs many complicated steps to get the goal. It contains vector design and construction, ES cells culture, colony selection, genotyping, embryo transfer, germ line transmission, and so forth. It is tedious and labor intensive work and difficult to approach. Among them, the vector design is the first and most important step as it has to have the entire concept regarding the animal model developed. In order to improve the designing of vector system, we developed reporter knock-in and gene knock-out system for the construction of vector as well as selection marker itself. With this progress, we can develop the knock-out mice more efficiently and accurately.
