Establishment of interaction between brain and peripheral immune system targeting immune biomarker in stroke mice

It is now increasingly clear that human stroke causes multi-organ system disease. Brain inflammation may lead to opposing local and systemic effects. Suppression of systemic immunity by the nervous system could protect the brain from additional inflammatory damage. However, it may increase the susceptibility to infection. The immune mechanisms involved in brain-peripheral interactions are still unknown. Therefore, we want to establish the interaction between brain and peripheral immune system targeting immune biomarker. Our strategies are followed; first, in different time points after tMCAO, we will analyze immune cell populations using flow cytometry in brain and peripheral immune tissues such as bone marrow, thymus, spleen, lymph nodes, blood and find systemic indicators which can be correlated to the severity of the stroke. Second, we have several immune biomarker candidates which may involve in the development of stroke e.g. complement C3, IL-4, IL-13, IL-6 etc. We will analyze their expression patterns in different time points after tMCAO and find their functions and mechanisms using overexpression or inhibition system. Third, we will establish co-culture systems between immune and brain cells which will mimic the brain inflammation system and inform us immune cell effect on brain cells. Finally, we will develop drugs which can regulate immune biomarkers and test whether they can be used for stroke therapeutics.
