뇌절편에서의 간질발생에 대한 전기생리학적 연구
초록
The in vitro slice provides a neuronal network with well-defined synaptic connections. The environment of the slice is also easily modified, and all the neurotransmitter systems are subject to pharmacological manipulation. This study describes a preparation that preserves the basic synaptic connections between somatosensory thalamus, nucleus reticularis thalami, and somatosensory cortex in vitro, and generates spontaneous thalamocortical rhythms of appropriate frequency, duration, and anatomical distribution when perfused with low Mg2+ medium. When perfused with Mg2+-free ACSF, thalamocortical slices exhibited two main types of epileptiform activity. The first are monotonic bursts that consist of an initial paroxysmal shift (PDS), followed by a period of 3-15 Hz synchronized bursting. The second are multiphasic bursts that have a tonic burst firing period and a late phase of clonic burst firing. These bursts are similar in frequency and duration to EEG recordings from human generalized seizures. Exploiting the advantages of in vitro preparations, the electrophysiological effects of ginsenosides on epileptiform activity were studied.
