The purpose of this study was to investigate the effect of lotus root ethanol extract(LREE)on human adipocyte differentiation into adipocyte, and expression of adipokines in differentiated adipocytes. Preadipocyte were cultured under differentiation conditions for 14 days in the various concentration (25, 50, 100, 200 mg LREE/ml) and differentiated adipocytes were also treated with LREE for 7 days. The morphological changes and degree of adiponegeisis of human adipocyte were measured by Oil-Red O staining assay. The culture supernatants were analyzed for adipokines (adiponectin, leptin, IL-6, and IL-8). It did not induce the dedifferentiation of adipocytes or inhibit fat accumulation in adipocytes. At concentrations of 100 and 200 mg/ml, LREE treatment resulted in significantly attenuated expression of adipogenic transcription factors such as peroxisome proliferator-activated receptor(PPARγ) and adipocyte marker genes such as glucose transporter 4(GLUT4) and leptin. The production of leptin, IL-6, and IL-8 was decreased and adiponectin was increased by LREE treatment for 7 days in IL-1β-stimulated differentiated adipocytes. These results indicate that LREE may modulate the expression of adipokines in adipocytes.
