Resolving conflicts among the experimental conclusions using TCGA and other public data base: a case study of Prss14/ST14 in breast cancers.

There exist confusing experimental conclusions on its role in tumorogenesis for a transmembrane serine protease, Prss14/suppression of tumor 14 (ST14).. While some transgenic expressions led to spontaneous adenoma/carcinoma, it also showed tumor suppression. The expression profilings of cell lines often was in contradiction to those in patients. By utilizing public databases, we investigated Prss14/ST14 expression and found that survivability is significantly poor in ER- breast cancer. The high expression of Prss14/ST14 regardless of the ratios to its inhibitors is the critical parameter. In clustering heatmaps, Prss14/ST14 is located closer to CDH2 than to CDH1 with patients, but, to CDH1 with cell lines. This apparent contradiction is not likely due to Prss14/ST14 expression in a cancer microenvironment. Coexpression analyses of known substrates revealed context dependent action. Therefore, it is important not to base exclusively on the cell line studies for target development.
