Effect of EGF-Endospray on Hemostatic Action in Gastric Bleeding Rabbit and Porcine Models

Gastrointestinal (GI) bleeding, one of the irritant problems in ulcer patients, is a serious symptom that occurs in any part of digestive tract. GI bleeding commonly causes from peptic ulcers. Epidermal growth factor (EGF) can accelerate regeneration of the mucous lesion, promote cell growth and exert beneficial effects by either reducing injury or accelerating repair at the wound area. In this study, we developed the ulcer-coating powder (EGF-endospray) to investigate the hemostatic action on gastric ulcer animal models. EGF-endospray was endoscopically designed and directly applied onto a mucosal resection (MR)-induced gastric bleeding focus in a rabbit model. In the rabbit model, EGF-endospray treatment exhibited a minor delayed bleeding time (104.3 sec), whereas the major delayed bleeding was observed in the non-treated control (548 sec) and the epinephrine injection (393.2 sec). In the micro-porcine model, EGF-endospray showed immediate hemostatic action and prolonged covering of the bleeding focus over 72 hrs. We believed that EGF was released from the ulcer-covering hydrogel, facilitates ulcer-healing and concurrently reduces re-bleeding. Histology proved mucosal thickness was more efficiently recovered in all EGF-endospray treated animals. The EGF-endospray could be new therapeutic endoscopic-based treatment for GI ulcer bleedings
