The monoterpene perillyl alcohol (POH) derived from orange peels has proven chemopreventive activity against rodent mammary cancer during the initiation phase as well as the promotion/progression phase with low toxicity. In this work, to isolate POH in Korean orange peel, the following steps were formulated. The extract from the powder of Korean orange peel was obtained by supercritical CO2 at the operating conditions of 50℃, 200 bar, and 6 CO2 kg/hr/kg sample. From the experiment results, most of POH was extracted within 14 hr. Then, the extracts were partitioned with acetonitrile/hexane, 80/20(vol.%). POH was presented in acetonitrile, and the non-polar materials was removed by hexane. The acetonitrile phase containing POH was further treated by open tubular chromatography to enhance the purity of POH in the extracts. Finally, RP-HPLC was applied to obtain POH with high purity. Separation was achieved on a μ-Bondapak C18 (3.9 × 300 mm, 10㎛) column and UV wavelength was fixed at 205 nm. The flow rate of mobile phase was 1 ml/min. The mobile phase for isolating POH from the extract consisted of water/acetonitrile, 70/30(vol.%) and water/methanol/acetonitrile, 60/20/20(vol.%).
