Poly Lactic-co-glycolic acid (PLGA) microparticles for reconstructive of tissue engineered articular cartilage

Introduction: Articular cartilage has a characteristic with limited capability self-repair. Tissue engineering has proved to be one of the most promising alternative for regenerative of articular cartilage. The purpose of this study is the use of PLGA microparticles with produced tissue engeering cartilage Materials and Methods: CHondrocytes were islated from New Zealand white rabbit knee articular cartilage. PLGA of 75:25 microparticles was prepared with a molecular weight of 100KD.After two passages, the chondrocytes were dynamic mixed on PLGA at densites of 1X107cells/ml. All samples were evaluated gross examination, colagen and GAG contents, histological and immunostaingn at 5, 6, 7, 8, 9 weeks cultured in vitro respectively. Native articular cartilage of rabbit was used as control group. Results: Whitish color, round shape and soft tissue as cartilage tissue was observed with gross examination at 5,6,7,8,9 weeks cultured. Collagen and GAG contents were reached 45.55% and 45.59% of native cartilage at 7 weeks of culturedd, respectively. Safranin-O and Alcian blue stains revealed similar to native cartilage tissue after 6 weeks of cultured. Homogenous distribution of collagne type II was observed at round of chondrocytes by immunostaining after 7weeks of cultured. Conclusion: We consider biodegradable PLGA microparticles as a useful produce for tissue engineering articular cartilage.
