Comparison of nitrogen sources for production of hCTLA4Ig in transgenic rice cell cultures

Human cytotoxic T-lymphocyte antigen 4-immunoglobulin (hCTLA4Ig) was produced using RAmy3D promoter induced by sugar starvation in transgenic rice cells. In this system, composition of culture medium, especially nitrogen sources, was found to be an important factor for the production of recombinant. N6 (CHU) medium containing inorganic nitrogen is conventionally used in rice cell cultures. Unlike inorganic nitrogen, uptake of amino acids can be more rapid compared to inorganic nitrogen. In this study, different types of nitrogen sources were applied in medium for the production of hCTLA4Ig. Relative viability of the cells cultured at day 3 in N6 medium was 3.9-fold higher than that in AA medium containing amino acids in production phase. Furthermore, production of hCTLA4Ig at day 9 in N6 medium was 1.54-fold higher than that in AA medium. In addition, supplementation of AA mixture (x10) to each medium led to enhanced production of hCTLA4Ig. In conclusion, N6 medium could enhance the level of hCTLA4Ig and cell viability in production phase. Moreover, the medium supplemented with AA mixture could provide synergistic effects on both the production of hCTLA4Ig and cell viability.
