Cytoprotective effects of leptin against MPP+ induced toxicity in SH-SY5Y Cells.

초록

The adipocyte-secreted hormone leptin is well known as a key hormone in energy homeostasis. The neurotoxin 1-methy1-4-phenylpyridinium(MPP+) mimics the neuropathology of Parkinson’s disease(PD) and likely kills dopaminergic neurons by inhibition of mitochondrial electron transfer and increase in level of oxidative stress. Recently, leptin was reported to involove in the modulation of neurotrophic or neuroprotective effects in vitro. Therefore , we investigated whether leptin protects neuronal cells against the MPP+-induced apoptotic death using SH-SY5Y neuroblastoma cell line, and the possible activation/inactivation of anti-apoptotic signaling pathways.(중략) In clnclusion, our results showed that leptin has cytoprotective effect against MPP+-induced cell death in SH-SY5Y cell, and that the underlying mechanisms for the leptin-mediated suppression of cell death is dependent on PI-3 kinase pathway.
