Reactive Oxygen Species Scavengers for Improving Cell Viability in Glycoengineered Rice Cell Cultures

Plant expression systems has been used for the production of therapeutic proteins. It has many advantages in terms of safety, production cost, scalability and post-translational modifications [1]. However, in cell culture conditions, plant cells encounter many stress conditions such as temperature, shear and salt stress. Since reactive oxygen species (ROS) occurs in all stress conditions, cell viability and target proteins could be negatively affected [2]. For maintaining ROS homeostasis under stress conditions, ROS should be controlled [3]. In this study, three different ROS scavengers (glutathione, melatonin, proline) were added in culture medium for reducing the formation of ROS in glycoengineered rice cells, which are known to be more sensitive compared to wild-type rice cells in culture. Cell viability was improved at day 3 by the addition of glutathione, melatonine and proline in glycoengineered rice cell cultures (21.45, 20.42 and 14.84%, respectively) compared to control conditions (11.64%). In conclusion, addition of ROS scavengers could be useful in glycoengineered rice cell cultures for improving cell viability by reducing stress.
