Pichia pastoris can utilize methanol as a carbon source and produce recombinant proteins under the control of the strong, tightly-regulated alcohol oxidase (AOX) promoter. High-density cell culture of P. pastoris is known to be easy, so scale-up is feasible. However, the presence of non-methanol carbon sources such as glycerol and glucose repressed fully the expression of AOX promoter. The expression level of recombinant proteins is also affected by the methanol induction conditions of culture. In this study, methanol induction conditions for high-level production of recombinant Hepatitis B surface Antigen (HBsAg) in P. pastoris were investigated. Addition of 2% acid mixture, 0.4% Pluronic F-68 or 0.01% oleic acid during methanol induction enhanced the production yield of HBsAg.
