Glucagon-Like-peptide 1 receptor polymorphism in Korean Type 2 diabetes

초록

Background : Glucagon-like peptide-1(GLP-1) is a hormone derived from the preproglucagon molecule that is secreted by intestinal L-cells and stimulates insulin secretion from betacells. The biologic actiion of GLP-1 are mediated by the GLP-1 receptor, which is 463 amino acids in size and has 7 transmembrane domains. Defects in insulin secretion are a common feature of type 2 diabetes and as a such the GLP-1 receptor is a candidate for contributing to the development clinically and genetically heterogenous disorder. Thus We investigated the association between single nucleotide polymorphism at position 260 in the GLP-1 receptor gene and type 2 diabetes Methods : The g.-260A>C polymorphism in the GLP-1 receptor exon 7 was determined using PCR-RFLP method in 424 type 2 diabetic patients and 248 healthy subjects Results : Allele frequencies of the 260A>C polymorphism of GLP-receptor gene were not different among Korean subjects with type 2 diabetes and control subjects. Also when we performed diabetic subjects into groups according to BMI(<25 Kg/m2, ??25 Kg/m2), diabetes onset age, allele frequencies of the 260A>C of GLP-1 receptor gene were not significantly different with control group. Conclusion : The 260A>C polymorphism in exon 7 of the GLP-1 receptor gene is not associated with type 2 diabetes in Koreans
