Taurine (2-aminoethan sulfonic acid) is the abundant amino acid in various mammalian tissues. Taurine shows multiple biological functions such as antioxidant, anti-stress, anti-inflammation, and antihypertensive effects. Obesity is international health problem caused by various environmental and genetic factors. To study the anti-obesity effect of taurine supplementation in diet-induced obesity rat model, five weeks-old male Sprague-Dawley rats were randomly divided into three groups for a period of 8 weeks (normal diet, N group; high fat diet, HF group; high fat diet + taurine, HFT group). Taurine was supplemented by dissolving in feed water (3% w/v), and the same amount of distilled water was orally administrated to N and HF groups. The serum total cholesterol level in the HF group was significantly decreased by taurine supplementation. The decreased serum adiponectin level in the HF group was significantly increased in the HFT group. In addition, after synthesis of new taurine-derivatives (TD) including taurine-glucose, taurine-galactose, taurine-ribose (TR), taurine-xylose (TX) and taurine lyxose (TL), we used human pre-adipocytes to investigate the effect of anti-obesity in vitro. Among these TRs, in vitro treatment with TR, TX, and TL resulted in inhibition of lipid accumulation. These results suggest that dietary taurine supplementation has beneficial effects on total cholesterol and adiponectin levels in high fat diet-induced obesity rats and various TDs has a potential to be developed as an effective anti-obesity reagent.
