초록
Transgenic Nicotiana tabacum suspension cells were cultivated in perfusion modes to produce recombinant human granulocyte macrophage-colony stimulating factor (hGM-CSF) continuously in a 5-L stirred tank bioreactor. During the perfusion culture for improving hGM-CSF productivity, cell density could be obtained up to 22.4 g/L which was 1.4-fold higher value compared to that of batch culture (16.1 g/L). The productivity of hGM-CSF could be maintained at 3 ?g/L/day until the end of cultivation with continuous addition of fresh medium and removal of spent medium. As a result of perfusion culture, 1.1 mg of extracellular hGM-CSF was accumulated in 26 L of total spent medium at the end of 16-day operation.
