Design and Synthesis of Ratiometric peptide probe for a Sensitive and Selective for Cu2+

초록

New peptide probe containing two pyrenes has been synthesized and studied as fluorescent chemo sensor for Cu2+. Two pyrenes were incorporated at each N and C terminal of tetramer peptide (Cys-Gly-Pro-Cys) inducing β-turn structure with amide bond formation. The synthesized peptide displayed a weak monomer and a strong dynamic excimer emission in acetonitirle and acetonitrle-buffer solution compared to typical pyrene emission. The peptide exhibited a ratiometric fluorescence change upon Cu2+ binding through enhancement of monomer emission and decrease of excimer emission with 1:2 stoichimetry and an isobestic point at 430nm. The fluorescent change mechanism was explained by conformational change of peptide after addition of Cu2+.
