Vesosomal drug delivery for controlled and sustained release of multiple components

Developments in drug delivery systems are promising for theragnostic drugs, including MOFs, micelles, peptides, hydrogels, and liposomes. Notably, liposomes? lipid-bilayer structures enclosing an aqueous core?offer biocompatibility, cellular uptake, and high drug capacity. However, limitations in loading diverse contents hinder controlled and prolonged release. To tackle this, we introduced vesosomes, larger liposomes containing smaller ones, like "peas-in-a-pod." The developed vesosomal drug delivery system incorporates a photosensitizer to generate reactive oxygen species (ROS), allowing the light-triggered release of encapsulated contents from ROS-sensitive inner liposomes while maintaining sustained release from ROSinsensitive liposomes. This innovative strategy holds the potential for advancing drug delivery and pioneers vesosomes as controlled drug carriers.
