Optimization of culture condition for cell growth in bubble column photobioreactors using central composite design (CCD)

Haematococcus pluvialis is the richest source of astaxanthin (3,3'-dihydroxy-?z?carotene-4,4'dione), which is a ketocarotenoid pigment with potential effects of cancer prevention, enhancer of immune response, and a free radial quencher. The effect of three factors (CO2 concentration, light intensity, flow rate) on cell growth of Haematococcus pluvialis were studied using central composite design. H. pluvialis UTEX 16 was used modified the modified Bold…s basal medium (MBBM) with optimized concentrations of Ca, Fe, Mg, Na, K, and trace elements in the medium. The culture temperature and pH have been maintained as 25oC and 6~7.5 respectively. The 0.4 L bubble-column photobioreactor were used for this experiments. The cells were inoculated at 1 ?e 104 cell/mL and experimental in fifteen bubble column photobioreactors with fed-batch cultivation for 8 days. The optimum values of these factors were obtained as follows: CO2 concentration: 3%, light intensity: 20 ?/m2/s, flow rate: 80 mL/min. The results showed that the operation condition that has been used in our lab was already an optimal condition and thus there was no significant different in the maximal cell density after the further optimization by CCD. However, these results identify the effect of the important factors on cell growth.
