초록
The prediction of the elution profile of single or mixed components, under non-linear condition, is important to optimize the experimental condition of preparative application. Reversde-Phase HPLC was used to get the elution profiles of thymidine, phenol or o-cresol. Frontal analysis(FA) and Pulsed-Input Method(PIM) were used to determine the parameters of the non-linear adsorption isotherm. For the Langmuir adsorption isotherm, the good agreement between the experimental and calculated profile by the two methods was observed. This implies that PIM characterized by simplicity and ease of experiments offers a better dynamic method to measure the single adsorption isotherm, compared to FA.
