Cloning and characterization of sodium-dependent glucose transporter 1 (SGLUT1) in Chinese fluk (Clonorchis sinensis)

The Clonorchis sinensis, fluke, lives in the common bile duct and gall bladder. C. sinensis is an anaerobic trematode as schistosomes, and C. sinensis utilizes exogenous glucose for energy production. C. sinensis transports external glucose in muscular tissues through the transporter protein. Glucose transporter consist of GLUT (facilitator) and SGLUT (sodium ion dependent). The purpose of this study was cloning and characterization of SGLUT in C. sinensis. A complementary DNA (cDNA) encoding a sodium-dependent glucose transporter 1 (SGLUT1) of C. sinensis was isolated from the adult C. sinensis cDNA library. The open reading frame of SGLUT1 cDNA consists of 1557 base pairs that encode a 519 -amino acid residue protein. Hydropathy analysis suggested that SGLUT1 possess 12 putative membrane spanning domains. When expressed in Xenopus oocytes, SGLUT1 mediated the transport of radiolabeled deoxy-D-glucose in a time- and sodium-dependent manner. No trans-uptakes of other small molecule transporters. It would be necessary kinetic and inhibition experiment to clarify the detail characteristics of this transporter protein and may give information for the molecular biological study of carbohydrate metabolism.
