Optimization of Concentration Ratio between Sucrose and Plant-derived Peptones for the Enhanced Production of hCTLA4Ig in Transgenic Rice Cell Cultures

Peptones have been used as the main nutritive components in cell culture media. They contain various peptides, amino acids, minerals, and other micronutrients that may serve as growth factors and help metabolic processes, thereby promote cell proliferation and the production of recombinant proteins. To enhance volumetric productivity, optimization of concentration ratio between plant-derived peptones (PPs) and sucrose is important in plant cell cultures. Therefore, it was necessary to investigate the effects of carbon and nitrogen sources for the improvement of recombinant protein production. Supplementation of defined medium with PPs was performed to enhance cell growth and the production of hCTLA4Ig in transgenic rice cell suspension cultures. Five combinations of sucrose and PPs were used to optimize the concentration of carbon and nitrogen sources. The higher concentration of sucrose enhanced cell growth but maximum hCTLA4Ig production was obtained with 3% (w/v) sucrose and 10 g/L PPs. Optimal concentration of PPs and sucrose was found to induce the production of hCTLA4Ig without media exchange. Therefore, this combination is useful for RAmy3D promoter system, which should be induced by sugar starvation and usually requires two-stage processes for the production of target proteins
