GENOMIC AND PHENOTYPIC CHARACTERISTICS OF STRAIN IMCC30063, BELONGING TO A NOVEL SPECIES OF POLYNUCLEOBACTER, ISOLATED FROM AN OLIGOTROPHIC FRESHWATER LAKE
초록
The genus Polynucleobacter belongs to the family Burkholderiaceae and is known to be one of the major lineages of free-living bacteria in diverse freshwater habitats, and many strains belonging to the genus have been cultivated. Phylogenetic analyses of 16S rRNA gene sequences showed that the Polynucleobacter strains cultured so far could be divided into four subclusters: PnecA, B, C, and D. We report here the cultivation and draft genome sequence of strain IMCC30063, a Polynucleobacter strain that is not affiliated with any of the four subclusters. Strain IMCC30063 was isolated from an oligotrophic freshwater lake by dilution-to-extinction method. After initial cultivation of extinction cultures, colonies of IMCC30063 were obtained and purified on R2A medium. Phenotypic characteristics of the strain were determined based on polyphasic approaches. The genome sequence of strain IMCC30063 was determined by Illumina sequencing and annotated by the RAST server.
