Production of Decursin Derivatives Using Root Cultures of Angelica gigas

In this study, production of decursin and decursinol angelate by root cultures of A. gigas was investigated. At first, analytical condition was optimized using HPLC to separate decursin and decursinol angelate. Rexchrome S5-100-ODS column was selected and 50% acetonitrile in water supported the best separation. Decursin and decursinol angelate showed different spectral characteristics respectively. Optimal UV wavelength for maximum sensitivity was determined to be 330 nm. For extraction from the cultured roots, 70% ethanol showed the best. Production of decursin was not noticed in root culture, but decursinol angelate was produced. The specific yield of decursinol angelate from cultured root was 0.1% (w/w). When methyl jasmonate was added into root cultures of A. gigas, significant elicitation of decursinol angelate was observed.
