Degradation of target protein by proteases is common in the suspension cultures of transgenic plant cells. Therefore, development of a new system is necessary for the recovery of unstable target proteins. In this study, the suspension cell culture of the transgenic Oryza sativa L. was carried out and the target protein, human cytotoxic-T-lymphocyte antigen 4-immunoglobulin (hCTLA4-Ig), was recovered by affinity column of recombinant Protein A (rProtein A). If rProtein A column was maintained at room temperature for 6 days, after the binding of hCTLA4-Ig without elution, most hCTLA4-Ig was degraded. For that reason, we cultured the cells in an air-lift bioreactor with continuous removal of medium from the culture. This study shows a new in situ recovery system for the transgenic rice cell cultures.
