In vitro sensitivity of Orientia tsutsugamushi isolated in Korea to doxycycline and clindamycin

We tasted the susceptibility of Orientia tsutsugamushi isolated in Korea to doxycycline and clindamycin. We have used the immunofluorescent antibody staining for the quantification of orientia in ECV304 cells. Because of oriential tendency to aggregate in ECV304 cells, we modified the IF staining method. After incubation with the antibiotic, the infected cells were disrupted by vortexing using glass beads to release the Orientia from the cells. The released orientia was re-infected the another ECV304 cells, and attached orientia was counted by IF staining after 3h for the counting total bacteria or after 2days for the counting viable bacteria. We found that the Orientia tsutsugamushi Boryong strain is susceptible to doxycycline and clindamycin.
