Construction of streptomycetes-E. coli shuttle vector containing bioluminescence system for potential environmental monitoring
초록
The bacterial luminescence system is highly valuable due to its application for an effective and simple environmental monitoring method. In this study, we constructed streptomycetes-E. coli shuttle vector containing bioluminescence system and evaluated its potential application for toxic compounds monitoring. The luxAB bioluminescence genes from Vibrio harveyi have been cloned into the streptomycetes-E. coli shuttle vector (named pESK004) and functionally expressed in streptomycetes. The recombinant Streptomyces lividans containing pESK004 exhibited the optimal luciferase activity at the optical density(OD600) of 0.4 and the aldehyde concentration of 0.005%. The luciferase activity from the recombinant luminescence streptomycetes was proportionally decreased against several heavy metals and chlorinated phenols. The EC50 (effective concentrations to decrease 50% of the luminescence prior to exposure) for Mercury and 2,4-dichlorophenol were measured at 2.2 ppm and 144 ppm, respectively.
