Lotus (Nelumbo Nucifera) seed has been reported to have various biological and phamacological properties. The purpose of the current study was to investigated whether lotus seed ethanol extract(LSEE) affects the differentiation of human preadipocytes into adipocytes and modulates the expression of adipokines. Preadipocyte were cultured under differentiation conditions for 14 days and the differentiated adipocytes were also treated with LSEE in the presence or absence of IL-1β(1 ng/ml) for 7 days. Oil Red O staining was performed to determine the lipid accumulation in human adipocytes. To elucidate the inhibitory mechanism of NSE on adipogenesis, peroxisome proliferator activated receptor γ(PPARγ), glucose transporter 4(GLUT4), and leptin were analyzed by RT-PCR. Lipid droplets produced during adipogenesis were significantly decreased in a dose-dependent manner by treatment of LSEE. Expression of major transcription factor (PPARγ), and adipocyte marker genes (GLUT 4 and leptin) of adipogenesis was decreased after treatment with LSEE. In addition, LSEE decreased production of IL-6 and IL-8 caused by IL-1β stimulation in a dose-dependant manner. These results suggest that lotus seed could be used as effective ingredients for obesity-related diseases.
