Electrophysiological study on epileptogenesis using acute in vitro brain slice model.
초록
1) Rats (Sprague Dawley, 4-6 weeks old) are decapitated under halothane anesthesia. The brains are rapidly removed and placed in ice-cold artificial cerebrospinal fluid (ACSF) containing (in mM) 126 NaCl, 3 KCl, 1.4 KH2PO4, 1.8 CaCl2·2H2O, 1.3 MgSO4, 23 NaHCO3, and 10 glucose, continuously bubbled with 95% O2/5% CO2. 2) Transverse hippocampal slices (400 mM thick) are prepared with a McIlwain tissue chopper (Hugo Sachs Electronik, March, Germany), and then incubated for at least 1 h in a chamber containing oxygenated ACSF before recording. A slice is transferred to a submerged recording chamber at 30℃ that is perfused with ACSF saturated with 95% CO2/5% O2 at a flow rate of 2-3 ml/min
