Production of Levofloxacin Using Immobilized Enzyme

Levofloxacin is L-form stereoisomer of ofloxacin. Its antimicrobial activity in superior to D-form. In this study, levofloxacin was produced by using porcine liver esterase(PLE) immobilized in polyacrylamide gel. PLE has enantioselective activity. The optimum gelling condition of monomer concentration was determined at 20% and optimum ratio of acrylamide and N,N'- methylenebisacrylamide was found to be 27.5:2.5. Immobilized enzyme could be reused 10 times. Regardless of repeated use, enzyme activity was maintained over 50% of free enzyme activity there was no reduction of enantioselectivity.
