We designed a preventive metastasis vaccine based on bioinformatics and the previously obtained data, and tested in the two breast cancer mouse models. Antigenic peptide vaccine, SP, contains the sequence derived from a membrane protease PRSS14 as a carcinoma marker. Although PRSS14 is a self-antigen, immunization of selected vaccine (conjugated with carrier) raised efficient antibody responses with or even without adjuvants. Vaccination efficiently abrogated the metastasis of the two independent breast cancer cell lines in the syngenic mouse strain hosts of different immunological predisposition, Th1 and Th2. Analyses of immune parameters in tumor bearing mice indicate that innate as well as antigen specific responses are involved. Cytotoxicity of the vaccine is not observed when vaccination was performed in Th2 type adjuvants. Therefore, these results suggest that the safe efficient vaccine approach is possible for preventing metastasis.
