소뇌 퍼킨지세포의 Na-Ca 교환 단백질의 생리적 기능
초록
: Variations in intracellular calcium activity ([Ca2+]i) play crucial roles in information processing in Purkinje neurons such as cerebellar long-term depression (LTD). Cerebellar LTD is expressed as a functional down regulation of AMPA receptor. Purkinje neurons have powerful systems to modulate [Ca2+]i, such as Ca2+-binding proteins, intracellular stores, Ca2+-ATPase and Na+-Ca2+ exchanger (NCX). The physiological role of NCX in Purkinje neurons is not well understood. NCX has special amino acid sequences for binding with caveolin. NCX-caveolin complex implicates that caveolin may be serve as a regulator of NCX activity. Thus, in this study, we investigated 1) the effects of NCX on the phosphorylation of AMPA receptor and the AMPA-activated responses 2) association of NCX with caveolin-1 by measurements of Ca2+ influx and efflux via NCX under inhibitory condition of caveolin-1 synthesis in cerebellar Purkinje neurons.
