Glutamine attenuates Zn (II) protoporphyrin IX-dependent methyl mercury toxicity in cultured mouse mesangial cells
초록
To clarify the effect of glutamine (Gln) on methyl mercury (MeHg)-induced renal toxicity, cytotoxicity and the change of heme oxygenase-1 (HO-1) gene expression wa studied using cultured mouse renal glomerular besangial cells (MMC). By employing RT-PCR and Western blotting analysis, we have examined the HO-1 induction in MMCs that were treated with MeHg. Using MTT assay we have also examined the cytoproteictive effect of HO-1 induction against the cytotoxicity caused by toxic dose of MeHg. In MMCs exposed to MeHg expression of HO-1 (mRNA and protein) was increased in a concentration and time-dependent manner. The increments of mRNA and protein expressions by MeHg were attenuated by the treatment of the cells with actinomycin D, an inhibitor of transcription, but no inhibitory effect of zinc protoprophyrin IX (ZnPP), an inhibitor of HO-1 activity, on MeHg-induced increment of HO-1 mRNA and protein expressions was observed. In cell viability test, the MeHg-induced cell death was enhanced by the treatment of Znpp. The decreased viabiliyt of the cell by NeHg was significantely recovered by the treatment of Gln with concentration dependent mammer. From these results, Gln have cytoprotictive effect that related with heme oxgensase-1 induction
