Epithin is a type II transmembrane serine protease that is highly expressed in epithelial cancer cells. It promotes angiogenesis, epithelial-mesenchymal transition (EMT), transendothelial migration and metastasis. 4T1 breast cancer cell line (stage III cancer model) highly express epithin in culture. While we maintained the 4T1 cell in IMDM, not a conventional media, we noticed that cells change morphology and epithin expression. 4T1 grown in IMDM lost their cell-cell contact like cells in post-EMT status, while cells grown in RPMI1640 maintained its original morphology like cells in pre-EMT status. The levels of epithin expression was diminished in IMDM-grown. In the assays of metastasis with IMDM-grown 4T1 in Balb/C, it failed to reveal any metastasis to the lung. To determine whether expression changed due to media composition, cells cultured in IMDM and RPMI1640 were compared. There exist small differences among salt, vitamin, and other nutrient between IMDM and RPMI1640, indicating that metabolic pathway might be involved in the regulation of epithin expression and metastasis. We now undertook a series of experiments to find factors that are responsible.
