Production of biopharmaceuticals from mammalian cell cultures has been steadily increased for the treatment of human diseases. Many researches have been performed to increase productivity in mammalian cell cultures. Chinese hamster ovary (CHO) cells are used widely for the industrial production of various recombinant therapeutic proteins. Storage-protein 2 (SP2) is a soluble protein derived from silkworm hemolymph. Molecular weight of SP2 is approximately 70 kDa and amino acid sequence is confirmed. In this study, we investigated the effect of SP2 on the recombinant CHO cells producing albumin-erythropoietin (Alb-EPO). Viable cell density and viability were increased by the addition of SP2. Also, it was found that the inhibition of aggregation as well as apoptosis was more effective than control. Aggregation rate was decreased by 20% at 2 mg/ml of SP2. Flow cytometer was used to confirm apoptosis. When the SP2 was added to media, apoptosis was also decreased. Productivity of Alb-EPO was increased 1.48-fold than the control. In conclusion, it was apparent that the addition of SP2 could enhance the productivity by reducing apoptosis.
