Development of in vitro 2D skin model for wound healing assay
초록
During wound healing process damaged skin is contracted and recovered by skin tissue components. The cellular behavior in skin tissue is changed by various environmental changes and affects wound healing time and remaining scars. In this research, we simply fabricated nano-wrinkled surfaces with biocompatible PDMS (Polydimethylsiloxane) polymer and analyzed the cellular behaviors, specifically cell migration, on the wrinkled surfaces during wound healing progress. We integrated a nano-wrinkled PDMS surface with microfluidic system which can mimic wound healing process. Nano-wrinkles were variously aligned along a microchannel from 0 to 90 degree. We acquired real time images of cell migration on the wrinkled pattern and found that simply fabricated nano-grooved surface guided the morphology and migration of cells, even the intercellular cytoskeleton structure. This result presents that cellular behavior can be controlled beneficially for wound healing by mechanical stimuli .We expect the 2D skin model integrated with various type of surface can be used as a standard assay platform for wound healing research.
