Analysis of Haematococcus pluvialis proteins associated with astaxanthin production

Haematococcus pluvialis is the richest sources of astaxanthin (3,3'-dihydroxy-?z?-carotene-4,4…-dione), a ketocarotenoid pigment, with potential effects of antioxidant. Cells are keeping in growth condition under optimal cell culture conditions while, by stresses, the cells enter in red cyst stage. Many different proteins may be induced in immature green and mature cyst cells. In this study, proteins of H. pluvialis were analyzed by native- and SDS-polyacrylamid electrophoresis (PAGE) and 2-dimensional electrophoresis. It showed different pattern of proteins. Native-PAGE resulted in some of different pattern on mature cyst cells compare to immature green cells. In SDS-PAGE analysis, proteins that have different molecular weight, 57 kDa, 47 kDa and 43 kDa, were induced in mature cells and were not detected immature green cells. In 2-D electrophresis analysis, quite unknown proteins were found in immature green and mature cyst cells. 2-D gel analysis will be studied more to understand the proteins structure associated with astaxanthin production by peptide mass fingerprinting (PMF), chemically assisted fragmentation (CAF)-MALDI sequencing.
