Changes of Stem Cell-specific Markers of Umbilical Cord Blood-derived Mesenchymal Stem Cells by Serial Passages

For the cell-based therapy, the primitive stem cells should be expanded without differentiation. Subcultures using trypsinization processes are required for the proliferation of stem cells that have adherent properties. It can cause the cells to lose their multipotency. In this study, the tendency of stem cell-specific surface marker of mesenchymal stem cells (MSCs) isolated from umbilical cord blood (UCB) by passaging was investigated. The growth rate of MSCs was measured by cell counting and immunophenotype was analyzed by fluorescence activated cell sorter (FACS) at passage 3, 5, 10, 15. Changes of CD73, CD90, CD105, CD166, which are positive markers of stem cells, were confirmed. These results can contribute to the development of ex vivo expansion condition of mesenchymal stem cells for clinical application.
