Transgenic plant cell cultures have been developed as a production system for producing therapeutic proteins. Human cytotoxic T-lymphocyte antigen 4-immunoglobulin (hCTLA4Ig) was produced by transgenic rice cell suspension cultures using RAmy3D promoter system. Since the cell wall inhibits the secretion of protein, the target protein, hCTLA4Ig can be caught inside cell wall or remained in intracellular space. Tween 20, a nonionic surfactant, has been proposed as a suitable permeabilizing agent for the permeabilization of plant cells in suspension cultures. To improve the release of hCTLA4Ig caught in cytoplasm and cell walls, Tween 20 was treated at the concentrations of 0.05, 0.1, 0.5, and 1.0% (v/v). Application of 0.5% Tween 20 enhanced the production of hCTLA4Ig by increased permeability. Therefore, the production and permeabilization of hCTLA4Ig could be enhanced by the optimal addition of Tween 20 in transgenic rice cell cultures.
