Cultivation and Draft Genome Sequence of Strain IMCC19396, a Freshwater Representative of the Acidobacteria, Isolated from Soyang Lake

The phylum Acidobacteria is one of the most abundantly distributed bacterial groups in terrestrial environments, but cultured isolates have been rarely reported from freshwater environments so far. Here we report the cultivation and genome characteristic of strain IMCC19396, a freshwater bacterial representative of the Acidobacteria. Strain IMCC19396 was obtained from a surface freshwater sample collected from the Soyang Lake through dilution-to-extinction culture using oligotrophic media. The 16S rRNA gene phylogeny revealed that strain IMCC19396 is affiliated with the subdivision 3 of Acidobacteria and is most closely related to Candidatus Solibacter usitatus Elin6076 (93.1% similarity) among cultured strains. Strain IMCC19396 did not form colonies on agar plates, but a stable growth was observed in the lake water-based oligotrophic liquid media, with a maximum cell density of ca. 107 ml-1. The draft genome sequence of IMCC19396 was determined by Illumina sequencing (250-bp paired-end). The genome sequence was assembled into 39 contigs, with a total length of 6.76 Mb (N50; 287 Kb). The DNA G+C content was 59.5 mol%. The genome of strain IMCC19396 has been found to possess 6,100 protein coding genes, among which genes for complete TCA cycle and RuBisCO were predicted.
