Bone marrow-derived mesenchymal stem cells (MSCs), also known as marrow stromal cells or multipotent adult progenitor cells (MAPCs), have multipotency to differentiate into developmentally different lineage cell types. To understand the molecular properties of MSCs such as stemness, self-renewal, and differentiation potential, several studies have investigated the gene expression profiles of MSCs. Although very informative, most of studies have limitation due to the heterogeneity of MSC populations. Recently we identified the human bone marrow-derived colony-forming fibroblastic cells established by subfractionation culturing method as MSC- or MAPC-like cells and described our protocol for obtaining homogenous clonal marrow stem cells (cMSCs) from bone marrow aspirates. A clone of our cMSC library, designated to D5-#2, was shown to have multipotency to differentiate into adipogenic, chondrogenic, osteogenic, neurogenic, and hepatogenic lineage cells, closely resembling MAPCs. The differential gene expression profiles of cMSCs during osteogenic or neurogenic differentiation were examined. The cMSCs were induced to osteogenic or neurogenic differentiation for 14 days, harvested to isolate RNA and genomic DNA, and then subjected to microarray and DNA methylation array, respectively. We analyzed the transcriptomes of cMSCs during osteogenic or neurogenic differentiation by correlating with DNA methylation array data. Here we describe the gene expression profiles obtained from our cMSCs during differentiation. These findings will help us expand our understanding of molecular property of cMSCs.
