Retinoic acid에 의한 SH-SY5Y세포의 신경세포 분화과정에서의 JNK신호전달계의 역할
초록
Neural differentiation is proceeded in multiple processes requiring various cellular factors. Here we demonstrate that c-Jun N-terminal kinase 1 (JNK1) plays an essential role in RA-induced neural differentiation of SH-SY5Y cells. Treatment of SH-SY5Y cells with 10 ? RA induced a strong activation of JNK1 within 10 min, and the immediate induction of the JNK1 activity returned to the basal level in an hour. The second surge of the JNK1 activity was observed around 1 day after treatment. Interestingly, both JNK1 and phospho-JNK was detected in the nucleus of cells during the immediate induction, while enriched phospho-JNK localization was detected in the neuritis during the delayed second activation period. In SH-SY5Y cell line overexpressing a dominant negative form of SEK1, the upstream kinase for JNK1, RA-induced differentiation of SH-SY5Y cells was reduced along with the inhibition of the immediate and late induction of JNK1 activation. These results suggest that JNK1 plays an essential role in RA-induced neuronal differentiation of SH-SY5Y cells.
