The Role of CD39 in mouse MSC-mediated T-cell suppression

Mesenchymal stem cells (MSCs) are known for their pluripotent differentiation ability and diverse immunomodulatory properties, especially for their capability to suppress T-cell responses. Among the factors involved in MSC-mediated immune regulation, CD39, a surface protein involved in adenosine production, has been extensively studied. However, the role of CD39 in MSCs has not yet been fully explored. In this study, we investigated the role of CD39 in mouse bone marrow-derived MSCs in regulating T cells. First, we observed varying expression levels of CD39 on different MSCs. Secondly, elevated CD39 expression enhanced T-cell suppression. This was indicated by a decreased production of pro-inflammatory cytokines and T-cell divisions. Third, we found that MSCs with elevated CD39 expression produced increased IL-6 expression. These findings highlight the importance of CD39 as a critical regulator of T cell responses by mouse MSCs and underscore the potential of CD39 as a biomarker for predicting MSC-mediated immunosuppression.
