The cleavage of poly(ADP-ribose) polymerase (PARP) by caspase-3 is an essential link with the apoptotic pathway in animal cells. In plant cells, however, there is no authentic evidence for the similar role of PARP during apoptosis. Using a heat shock(HS)-induced apoptosis system of tobacco cells, it was found that DNA laddering appeared only after 20 h recovery following the HS. When cells were preincubated with 4 mM of 3-aminobenzamide (3-AB) or 2-4 mM of nicotinamide (NIC), as specific inhibitors of PARP before HS treatment, apoptotic cell death was significantly reduced and cell growth was recovered. Cell viability was the best at day 6. In addition, when cells were preincubated with NIC before camptothecin treatment for induction of apoptosis, cell growth and viability was recovered relatively. However, addition of NIC without HS or camptothecin treatment reduced growth rate.
