Cultivation and genomic analyses of three new Flavobacteriaceae strains, isolated from a marine solar saltern
초록
Three novel f!avobacterial sσains that are gram-stain-negative, non-motile, and orange-colored were isolated from a marine solar saltem, and were designated IMCC2192T ’ IMCC2184, and IMCC2644. Optimal growth of the three strains were obseπed at 25-30oC, pH 6.0-7.5, and with 1-4% (w/v) NaC1. The DNA G+C content of the strains were 35.9-36.0 mol%. For all the three strains, major respiratory quinone was menaquionone-6 (MK-6), and iSO-C' 5: ' G, iSO-C' 5:0 and iso-C 17:o 30H were shown to be predominant cellular fatty acids. Aminolipids, phospholipids, and phosphatidylethanolamine were major polar lipids of three strains. Phylogenetic analysis based on 16S rRNA gene sequences indicated that the strains belonged to the family Flavobacteriaceae and formed a distinct lineagc that was independent of closely related genera: Winograds양'ella (95 .1-92.8%), Flavirhabdus (94.2-94.3%), Bizionia (92.8-95.1 %), and Corallibacter (93.9-94.0%). Average nucleotide identity (ANI) values based on whole genome sequence data were >98% between the strains IMCC2192T and IMCC2644 al1d DNA-DNA relatedness based on hybridization was >90% between the strains !MCC2192T and IMCC2184, which indicated that the three strains belong to the same genomics species. The genome size of strains IMCC2192 T and IMCC2644 were 2.9 Mb and 2.8 Mb, respectively, and the two genomes shared general genomic properties of the Flavobacteriaceae. 8ased on the above results, including phylogenetic analysis and several phenotypic characteristics, strain αtlCC2192 , was considered to represent a novel genus within the family Flavobacteriaceae, for which the name Aurantibacter yeongjongensis gen. nov., sp nov. is proposed. The type strain is IMCC2192T, (=KACC18774').
