Efficacy test of Methyl Methoionine Sulphonium Chloride on indomethacin induced gastric mucosal damage in rats.

Methyl-methionine sulphonium chloride (Vitamin U) is a vitamin-like active substance. Its deficiency is thought to be a possible cause of gastric ulcers. MMSC has long been shown effective for treating peptic ulcers, but the mechanisms for its action are poorly understood in vivo. Here we explored anti-ulcer effects of MMSC on Indomethacin (Non steroidal anti-inflammatory drugs) Induced gastric mucosal damage in rats. We divided the animals in 4 groups as follows: (1) 25 mg/kg indomethacin, (2) 25 mg/kg indomethacin + 32 mg/kg MMSC, (3) 25 mg/kg indomethacin + 125 mg/kg MMSC, (4) 25 mg/kg indomethacin + 500 mg/kg MMSC. In groups 2, 3 and 4, indomethacin was given at 30 minutes after MMSC administration. 25 mg/kg indomethacin was given to all rats (n=10) by oral gavage in 3% sodium bicarbonate. Seven hours after, the entire rats were sacrificed and confirmed gastrointestinal erosions and ulcers. Pretreatment with MMSC (32, 125 and 500 mg/kg) at 30 minutes before administration of indomethacin resulted in a decrease in gastric damage areas. Among them, 32 mg/kg MMSC was the most effective against Indomethacin Induced gastric mucosal damage. We had similar patterns of result in mucosal concentrations of proinflammatory cytokines analysis, superoxide dismutase (SOD), malondialdehyde (MDA) and cyclooxygenase-2(COX-2) levels. Our data suggests, MMSC accomplishes the protective effect on indomethacin-induced gastric mucosal damage in a dose independent manner.
