ISOLATION OF MMS RESISTANT YEAST MUTANTS TO SEARCH FOR THE GENES THAT PROTECT CELLS FROM TOXICANTS.

Environmental DNA damaging agents are toxic to the cell. They cause wide-ranging cellular response including cell cycle alteration, metabolic changes, mutation and even cell death. Cells actively protect themselves from the damaging agents by eliminating toxic chemicals and recover from damage by damaged cellular components repair including damaged DNA. Previously, we showed that effect of MMS on cell survival and mutagenesis. Recently, we observed that damaged DNA repair gene mutation leads to decreased lifespan in yeast. In order to identify genes that might increase yeast life span in the presence of DNA damage, we attempted to isolate genes that increase or decrease MMS sensitivity using yeast insertional deletion library. Several genes that were previously known to be un-related to MMS sensitivity were identified to increase or decrease MMS sensitivity. Currently, we are confirming the effect of each gene on MMS sensitivity by investigation of the characteristics of targeted-gene deletion mutants.
