Ploymorphisms of XRCC1 codon 399 affect progression-free survival in non-small cell lung cancer.

Since the XRCC1 399Gln variants are common and the base excision repair is particularly important for repairing oxidative base damage by ionizing radiation exposure, the genetic polymorphism may have clinical significance in the management of lung cancer patients. We hypothesized that XRCC1 399Gln variant contributes to sensitivity to ionizing radiation treatment and is inversely associated with time to progression and survival. Materials and methods One hundred and thirth-six lung cancer patients were recruited at Asan Medical Center from 2000 to 2003. The elifibility criteria were stage I to ⅣNSCLC and ECOG performance score 0-2. We determined the genotypes of the XRCC1 genes by PCR-RFLP. The PCR products were digested with MspI and analyzed in a 3 % Metaphor gel. Kaplan-Meier wurvival curves and the log-rank test were used to analyze the genotype effect on progression-free survival. The hazard ratio adjusted for age, sex, and other potential confounders was calculated by Cox-proportional hazard moded. Results Number of patients with XRCC1 399Arg homozygote was 70and that with XRCC1 399Gln allele was 63.
